A decade of learning about the HIV reservoir: new insights into latent infection.

چکیده

One longstanding question is how cells become latently infected. In 2007, Suha Saleh (Monash University, Australia) and colleagues published an article that provided one of the first pieces of the puzzle. They demonstrated that when resting CD4+ T cells are incubated with certain chemokines and then infected with HIV, many copies of the virus are able to integrate into the cells’ genomes. [1] Sharon Lewin (University of Melbourne), the senior author of the article, observes that this study “was the first to show that latency

Human immunodeficiency virus type 1 can establish latent infection in resting CD4+ T cells in the absence of activating stimuli.

Resting CD4(+) T cells are the best-defined reservoir of latent human immunodeficiency virus type 1 (HIV-1) infection, but how the reservoir is formed is unclear. Understanding how the reservoir of latently infected cells forms is critical because it is a major barrier to curing HIV infection. The system described here may provide an in vitro model of latent HIV-1 infection in resting CD4(+) T ...

The Latent Reservoir for HIV-1: How Immunologic Memory and Clonal Expansion Contribute to HIV-1 Persistence.

Combination antiretroviral therapy (ART) for HIV-1 infection reduces plasma virus levels to below the limit of detection of clinical assays. However, even with prolonged suppression of viral replication with ART, viremia rebounds rapidly after treatment interruption. Thus, ART is not curative. The principal barrier to cure is a remarkably stable reservoir of latent HIV-1 in resting memory CD4(+...

The host cell side of latent HIV-1 infection

The ability of HIV-1 to establish an extremely stable latent viral reservoir in the CD4 + memory T cell population prevents viral eradication with the currently available antiretroviral drugs. A viral reservoir consisting of only 10 5 latently HIV-1 infected T cells could take more than 60 years to dissipate, making natural eradication during the lifetime of a patient impossible [1]. To achieve...

The latent HIV-1 reservoir in patients undergoing HAART: an archive of pre-HAART drug resistance.

Recent studies on patients with a history of pre-HAART drug resistance, but currently on a successful regimen, provided new insights into the dynamics of the latent cellular viral reservoir. Results indicated that the latent reservoir is an archive, composed of a mixture of wild-type and drug-resistant strains. The studies showed that, even after years of successful HAART, the wild-type viral s...

HIV Can Establish Latency by Direct Infection of Resting CD4+ T Cells

HIV establishes a latent reservoir in a small pool of resting CD4+ T cells early in the infection of a new host. This viral reservoir has a very slow rate of decay and it is resistant to anti-retroviral therapy and the immune surveillance. This reservoir poses a significant obstacle to the eradication of virus in an infected individual. It is thus of importance to understand how this reservoir ...
